Description

Spleen tyrosine kinase (SYK) is a 70-80 kDa cytoplasmic non-receptor protein tyrosine kinase, a member of the protein kinase superfamily. SYK is widely expressed in hematopoietic cells specially B lymphocytes, where it couples immunoglobulin receptors and is also expressed in fibroblasts, and endothelial cells. SYK plays an important role in transmitting signals from a variety of cell surface receptors and in signaling events mediating cellular proliferation, differentiation, and phagocytosis (1). SYK expression is up regulated in T cell lymphoma, thus playing an important role in tumorigenesis.

SYK is involved in intracellular signaling of the multi-chain immune receptors, including B cell receptor (BCR), zeta chain of T-cell receptor (TCR), FcR and integrins, which contains the immune-receptor tyrosine-based activation motif (ITAM). The ITAM adaptors or ITAM-containing receptor tails mediate signaling from B- and T-cell receptors, Fc receptors and many C-type lectins. SYK also mediates cellular adhesion, innate immune recognition, osteoclast maturation, platelet activation and vascular development.

SYK is activated by C-type lectins and integrins. Inhibition of SYK has a potential to regulate B-cell mediated inflammatory diseases such as SLE (2).SYK inhibitor, fostamatinib, exerts potent therapeutic efficacy against autoimmune and allergic diseases such as rheumatoid arthritis (RA), bronchial asthma and idiopathic thrombocytopenic purpura (ITP) (3). SYK blockade prevents the development of skin and kidney lesions in lupus-prone mice. Inhibition of SYK kinase activity impinges on various innate and adaptive immune responses. Selective SYK inhibitors are used for the treatment of inflammation-related and autoimmune-related disorders (4).

SYK- selective antibodies were generated against a peptide taken from the human SYK protein spanning aa from 240-330. The SYK -selective antibodies are affinity purified on an immobilized antigen based affinity matrix, the isolated antibodies were then stabilized in antibody stabilization buffer for long-term storage. The anti- SYK -selective antibodies are fully characterized for applications in western blotting and ELISA at the recommended dilutions
